{"id":145,"date":"2023-02-16T01:52:32","date_gmt":"2023-02-15T17:52:32","guid":{"rendered":"http:\/\/39.105.184.3\/?p=145"},"modified":"2025-12-21T01:06:34","modified_gmt":"2025-12-20T17:06:34","slug":"%e5%9f%ba%e4%ba%8ecrispr-cas9%e5%92%8ccre-loxp%e7%9a%84%e6%88%90%e5%b9%b4%e5%b0%8f%e9%bc%a0%e5%9f%ba%e5%9b%a0%e6%95%b2%e9%99%a4","status":"publish","type":"post","link":"https:\/\/fanyiming.life\/?p=145","title":{"rendered":"\u57fa\u4e8eCRISPR\/Cas9\u548cCre-LoxP\u7684\u6210\u5e74\u5c0f\u9f20\u57fa\u56e0\u6572\u9664"},"content":{"rendered":"\n<p>\u5bf9\u5c0f\u9f20\u8fdb\u884c\u57fa\u56e0\u6572\u9664\u662f\u7814\u7a76\u76ee\u7684\u57fa\u56e0\u529f\u80fd\u7684\u5e38\u7528\u65b9\u6cd5\u3002\u4f20\u7edf\u7684\u3001\u4ece\u53d7\u7cbe\u5375\u6216\u80da\u80ce\u65f6\u671f\u8fdb\u884c\u57fa\u56e0\u64cd\u4f5c\uff0c\u6765\u83b7\u5f97\u57fa\u56e0\u6572\u9664\u5c0f\u9f20\u7684\u65b9\u6cd5\u5f80\u5f80\u8017\u65f6\u957f\u8fbe\u51e0\u5e74\u3002AAV\uff08\u817a\u76f8\u5173\u75c5\u6bd2\uff09\u9012\u9001\u7cfb\u7edf\u7ed3\u5408CRISPR\/Cas9\u53ef\u4ee5\u5b9e\u73b0\u5bf9\u6210\u5e74\u5c0f\u9f20\u7684\u57fa\u56e0\u6572\u9664\uff0c\u4ece\u800c\u5927\u5927\u7f29\u77ed\u83b7\u5f97\u57fa\u56e0\u6572\u9664\u5c0f\u9f20\u7684\u65f6\u95f4\u3002AAV\u8fd8\u5177\u6709\u4f4e\u514d\u75ab\u539f\u6027\u3001\u4e0d\u540c\u8840\u6e05\u578b\u5b9e\u73b0\u9776\u5411\u4e0d\u540c\u7ec4\u7ec7\u6216\u5668\u5b98\u7684\u4f18\u70b9\u3002<\/p>\n\n\n\n<p>\u4e0e\u6b64\u540c\u65f6\uff0cAAV\u9012\u9001\u7cfb\u7edf\u6709\u4e00\u4e2a\u56fa\u6709\u7f3a\u9677\uff0c\u5176\u8f7d\u4f53\u6240\u80fd\u5bb9\u7eb3\u7684\u63d2\u5165\u5e8f\u5217\u6700\u5927\u53ea\u67094.7Kb\u3002\u4f7f\u7528CRISPR\/Cas9\u8fdb\u884c\u57fa\u56e0\u6572\u9664\uff0c\u9700\u8981\u5c06\u542b\u6709Cas9\u7f16\u7801\u5e8f\u5217\u548csgRNA\u7684\u8f7d\u4f53\u8fdb\u884c\u9012\u9001\u3002\u7136\u800c\uff0c<em>Streptococcus pyogenes<\/em> (SpCas9) \u548c gRNA together\u5c31\u5360\u4e864.2kb\uff0c\u5bf9AAV\u7684\u5bb9\u91cf\u4ea7\u751f\u4e86\u6311\u6218\u3002\u56e0\u6b64\uff0c\u5982\u4f55\u5bf9Cas9\u8fdb\u884c\u6709\u6548\u7684\u9012\u9001\u4e00\u76f4\u662fCRISPR\/Cas9\u5728\u52a8\u7269\u4f53\u5185\u53d1\u6325\u4f5c\u7528\u7684\u96be\u70b9\u3002\u8fc7\u53bb\u7684\u65b9\u6cd5\u5305\u62ec\uff1a\u5728\u4e0d\u540c\u7269\u79cd\u4e2d\u5bfb\u627e\u5e8f\u5217\u66f4\u77ed\u66f4\u6709\u6548\u7684Cas9\u3001\u5c06Cas9\u5206\u6bb5\u8868\u8fbe\u5728\u4e24\u4e2aAAV\u8f7d\u4f53\u7b49\u3002<\/p>\n\n\n\n<p>\u5317\u5927\u738b\u6653\u4f1f\u8bfe\u9898\u7ec4\u4e8e2021\u5e749\u6708\u5728STAR Protocols\u6742\u5fd7\u53d1\u8868\u4e86\u4e00\u7bc7\u6587\u7ae0\u8bb0\u5f55\u4e00\u79cd\u7ed3\u5408\u4e86Cre-LoxP\u548cCRISPR\/Cas9\u3001\u7528AAV\u9012\u9001\uff0c\u5b9e\u73b0\u5bf9\u6210\u5e74\u5c0f\u9f20\u8fdb\u884c\u7ec4\u7ec7\u7279\u5f02\u6027\u57fa\u56e0\u6572\u9664\u7684\u65b9\u6cd5\u3002<\/p>\n\n\n\n<p>\u539f\u7406\u5927\u81f4\u4e3a\uff1a<\/p>\n\n\n\n<figure signal\uff0cspCas9\u5f97\u4ee5\u8868\u8fbe\uff0c\u4e0eCre\u4e00\u540c\u8f6c\u5bfc\u5165\u52a8\u7269\u7684\u8fd8\u6709sgRNA\uff0c\u4e8e\u662f\u5b9e\u73b0\u5728\u809d\u810f\u4e2d\u5bf9\u8be5sgRNA\u9776\u5411\u7684\u7247\u6bb5\u8fdb\u884c\u5207\u5272\u3002spCas9\u540e\u9762\u8fd8\u63a5\u4e86GFP\u65b9\u4fbf\u5b9e\u9a8c\u8005\u89c2\u5bdfCre\u9176\u662f\u5426\u6210\u529f\u53d1\u6325\u4f5c\u7528\u3002<\/p>\n\n\n\n<hr
