H\u5bf9\u6742\u4ea4\u94fe\u4e2dRNA\u7684\u964d\u89e3\u5b9e\u73b0\u6572\u964d(RNase H-dependent)\uff0c\u540e\u8005\u4e0d\u4f9d\u8d56\u4e8eRNaseH\uff0c\u4f9d\u9760\u4e0e\u9776RNA\u7d27\u5bc6\u7ed3\u5408\u540e\u7684\u7a7a\u95f4\u4f4d\u963b\u6291\u5236\u7ffb\u8bd1\u548c\u526a\u63a5\u5b9e\u73b0\u6572\u964d(RNase H-independent)\u3002<\/p>\n\n\n\n<h1 class=\"wp-block-heading\">FANA<\/h1>\n\n\n\n<p>\u4e3a\u4e86\u66f4\u597d\u7684\u8bf4\u660eFANA\u7684\u7279\u70b9\uff0c\u5148\u4ecb\u7ecd\u548c\u5b83\u76f8\u5173\u7684\u5176\u4ed6\u79cd\u7c7b\u7684ASOs\u3002<\/p>\n\n\n\n<p>\u5df2\u77e5DNA-RNA hybrid\u53ef\u4ee5\u88abRNase H\u8bc6\u522b\uff0c\u5bf9\u6742\u4ea4\u94fe\u4e2d\u7684RNA\u8fdb\u884c\u5185\u5207\u3002\u5229\u7528\u8fd9\u4e00\u539f\u7406\u7684ASOs\u79f0\u4e3aRNase H-dependent ASOs. \u8fd9\u4e00\u7c7bASO\u53d1\u6325\u4f5c\u7528\u4e3b\u8981\u9700\u8981\u6ee1\u8db33\u4e2a\u6761\u4ef6\uff1a<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>High RNA binding affinity<\/li>\n\n\n\n<li>Nuclease resistance<\/li>\n\n\n\n<li>RNase H activation<\/li>\n<\/ul>\n\n\n\n<p>\u5373\uff0c\u5728\u4fdd\u8bc1\u4e0e\u9776RNA\u9ad8\u4eb2\u548c\u529b\u7684\u540c\u65f6\uff0c\u80fd\u591f\u9ad8\u6548\u6fc0\u6d3bRNase H\u7684\u5185\u5207\u6d3b\u6027\uff0c\u540c\u65f6\u4fdd\u8bc1\u81ea\u8eab\u80fd\u591f\u62b5\u6297\u6838\u9178\u9176\u964d\u89e3\uff0c\u4fdd\u6301\u8f83\u957f\u7684\u534a\u8870\u671f\u3002\u4e3a\u4e86\u5b9e\u73b0\u8fd9\u4e9b\u6761\u4ef6\uff0c\u9700\u8981\u5728\u5929\u7136DNA\u7684\u57fa\u7840\u4e0a\u8fdb\u884c\u5316\u5b66\u4fee\u9970\u3002<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">\u786b\u4ee3\u78f7\u9178\u916f\u952e<\/h2>\n\n\n\n<figure class=\"wp-block-image\"><img loading=\"lazy\" decoding=\"async\" decoding=\"async\" class=\"wp-block-list\">\n<li>\u4f7fASO\u6784\u8c61\u66f4\u63a5\u8fd1RNA\uff0c\u4ece\u800c\u6781\u5927\u589e\u5f3aASO\u7684\u5bf9target RNA\u7684binding affinity.<\/li>\n<\/ul>\n\n\n\n<p>\u7f3a\u70b9\uff1a<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>\u65e0\u6cd5\u6fc0\u6d3bRNase H\uff08\u56e0\u4e3a\u6742\u4ea4\u94fe\u6784\u8c61\u66f4\u63a5\u8fd1RNA-RNA hybrids\u800c\u975eRNase H\u9700\u8981\u7684DNA-RNA H\u7684\u7f3a\u70b9\uff0c\u73b0\u5728\u901a\u5e38\u53ea\u5728\u4e24\u6bb5\u51e0\u4e2abases\u52a02\u2019-O-MOE\u4fee\u9970\uff08wings\uff09\uff0c\u4e2d\u95f4\u75596~10\u4e2abases\u4f5c\u4e3aRNase H\u5207\u5272\u4f4d\u70b9\uff08gap\uff09\uff0c\u79f0\u4e3a\u201dgapmer\u201d\u6280\u672f\u3002<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">FANA<\/h2>\n\n\n\n<figure class=\"wp-block-image\"><img \u7136\u800c\uff0c\u5728FANA\u4e2d\u5f15\u5165\u786b\u4ee3\u78f7\u9178\u916f\u952e\u4f1a\u5728\u4e00\u5b9a\u7a0b\u5ea6\u4e0a\u964d\u4f4eRNase H activation\u80fd\u529b\u3002\u6240\u4ee5\uff0cFANA\u5728\u88ab\u7528\u4e8eASOs\u4e5f\u88ab\u8bbe\u8ba1\u6210PS-(FANA-DNA-FANA) chimeric gapmers\u6216Altimers\u7684\u5f62\u5f0f\u3002<\/p>\n\n\n\n<figure class=\"wp-block-image\"><img loading=\"lazy\" decoding=\"async\" decoding=\"async\"
