interconnected network of transcription-dependent and tunable microgels (biorxiv.org)<\/a><\/p>\n\n\n\n<p>\u79d1\u5b66\u95ee\u9898\uff1a\u7ec6\u80de\u6838\u5185\u7684\u5fae\u73af\u5883\u662f\u600e\u6837\u7684<\/p>\n\n\n\n<p>\u63d0\u51fa\u5047\u8bbe\uff1aSAF-A\u548cnuclear RNA\u5f62\u6210\u4e00\u79cd\u80f6\u72b6\u7f51\u7edc\u7ed3\u6784\uff0c\u6587\u4e2d\u6210\u4e3a&#8221;microgel&#8221;\u3002microgel\u5185\u90e8\u86cb\u767d\u8d28\u7684\u6269\u6563\u901f\u7387\u66f4\u4f4e\uff0c\u800cmicrogel\u591a\u7684\u5730\u65b9\u7684\u67d3\u8272\u8d28\u66f4\u677e\u6563\u3001\u8f6c\u5f55\u66f4\u6d3b\u8dc3\u3002microgel\u672c\u8eab\u53ef\u4ee5\u88ab\u4e00\u79cd\u6838\u9178\u5185\u5207\u9176XRN2\u964d\u89e3\u3002<\/p>\n\n\n\n<p>\u4e3b\u8981\u5b9e\u9a8c\uff1a<\/p>\n\n\n\n<p>1\uff09\u901a\u8fc7\u8d85\u5206\u8fa8\u663e\u5fae\u955c\u89c2\u5bdf\u5230SAF-A\u548cnuclear \/><\/figure>\n\n\n\n<p>\u56feG\u5bf9\u4ea4\u8054\u540e\u5bf9SAF-A\u505aWB\uff0c\u5f53\u03b1-amanitin\u6291\u5236\u8f6c\u5f55\uff0c\u540c\u65f6\u5b58\u5728XRN2\u65f6\uff0cSAF-A\u66f4\u591a\u662fmonomer.<\/p>\n\n\n\n<p>\u6700\u540e\u8fd8\u6709\u4e00\u4e9bfigure\u7814\u7a76\u4e86micro\u5bf9\u67d3\u8272\u8d28\u201c\u53bb\u538b\u7f29\u201d\u7684\u529f\u80fd\u3002<\/p>\n\n\n\n<p>\u603b\u7ed3\uff1a\u672c\u6587\u7814\u7a76\u63ed\u793a\u4e86RNA\u548cSAF-A\u76f8\u4e92\u4f5c\u7528\u5f62\u6210\u7684\u5fae\u51dd\u80f6\u5728\u7ec6\u80de\u6838\u5185\u7684\u91cd\u8981\u4f5c\u7528\u3002\u901a\u8fc7\u9ad8\u5206\u8fa8\u7387\u6210\u50cf\u548c\u5206\u5b50\u52a8\u529b\u5b66\u6a21\u62df\uff0c\u4f5c\u8005\u53d1\u73b0\u8fd9\u4e9b\u5fae\u51dd\u80f6\u4fc3\u8fdb\u67d3\u8272\u8d28\u53bb\u538b\u7f29\uff0c\u4ece\u800c\u8c03\u63a7\u57fa\u56e0\u8868\u8fbe\u3002\u8fd9\u4e00\u673a\u5236\u901a\u8fc7RNA\u964d\u89e3\u9176XRN2\u7684\u8c03\u63a7\u8fdb\u4e00\u6b65\u5f97\u5230\u4e86\u9a8c\u8bc1\u3002<\/p>\n\n\n\n<p><\/p>\n\n\n\n<p>\u4e00\u4e9bQ&amp;A.<\/p>\n\n\n\n<p>1\uff09\u76ee\u5149\u600e\u4e48\u8f6c\u79fb\u5230XRN2\u4e0a\u7684\uff1f<\/p>\n\n\n\n<p>\u7b54\uff1a\u901a\u8fc7BioID\u6280\u672f\u53d1\u73b0XRN2\u662fSAF-A\u7684proximal proteins. \u7ed3\u5408XRN2\u662f\u4e00\u79cd\u6838\u9178\u5185\u5207\u9176\uff0c\u4e8e\u662f\u8054\u60f3\u5230\u5b83\u662f\u5426\u80fd\u901a\u8fc7\u964d\u89e3nuclear RNA\u6765\u8c03\u63a7microgel.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>We reasoned that proteins important for RNA turnover were likely to be spatially located in the vicinity of the nucleoplasmic network and microgels, so BioID74 (proximity-dependent biotin identification) was used to identify proteins in the vicinity of the SAF-A, by tagging the protein on the N-terminus with BirA.
